Choline dendrimers as generic scaffolds for the non-covalent synthesis of multivalent protein assemblies.
A modular self-assembly strategy is presented that allows the non-covalent synthesis of multivalent protein dendrimers using the strong interaction between choline-functionalized dendrimers and the choline binding protein C-LytA. Choline dendrimers displaying fusion proteins of C-LytA and the collagen binding protein CNA35 represent attractive multivalent targeting ligands for collagen imaging.
متن کاملOrthogonal ligation strategies for peptide and protein.
This review focuses on the concept, criteria, and methods of an orthogonal amide ligating strategy suitable for syntheses of peptides, peptide mimetics, and proteins. Utilizing unprotected peptides or proteins derived from chemical or biosynthetic sources, this ligation strategy has been shown to be general and exceptionally mild. Its orthogonality in ligating two unprotected segments with free...
متن کاملOne-pot native chemical ligation of peptide hydrazides enables total synthesis of modified histones.
One of the rising demands in the field of protein chemical synthesis is the development of facile strategies that yield the protein in workable quantities and homogeneity, with fewer handling steps. Although the native chemical ligation of peptide hydrazides has recently been shown to be useful for the chemical synthesis of proteins carrying acid-sensitive modification groups, previous hydrazid...
متن کاملProtein Assembly Using the Staudinger Ligation
Introduction New methods are facilitating the total chemical synthesis of proteins. In particular, the chemical ligation of synthetic peptides provides a convergent route to proteins. Currently, the most common ligation method is “native chemical ligation” [1]. In native chemical ligation, the thiolate of an N-terminal cysteine residue of one peptide attacks the C-terminal thioester of a second...
متن کاملProtein synthesis by native chemical ligation: expanded scope by using straightforward methodology.
The total chemical synthesis of proteins has great potential for increasing our understanding of the molecular basis of protein function. The introduction of native chemical ligation techniques to join unprotected peptides next to a cysteine residue has greatly facilitated the synthesis of proteins of moderate size. Here, we describe a straightforward methodology that has enabled us to rapidly ...
